The TDSC provides basic histology services including processing tissues and cell pellets, embedding, and cutting both frozen and paraffin-embedded samples.
We can provide unstained slides, rolls, H&E's and some special stains.
The TDSC performs both single and multiplex immunohistochemistry and immunofluorescence. We have an extensive menu of routinely used antibodies and provide a workup
service for new antibodies. We have experience with multiple techniques and chromagens allowing us to optimize for the best results.
The TDSC has a Leica Bond III autostainer. This instrument enables us to run large studies with minimal variability and workup new tests for the clinical lab.
The TDSC works closely with the clinical IHC lab and its Medical Director to ensure that the best clones and most up-to-date assays are in use.
Tissue Microarray Construction
The TDSC can construct high and low-density arrays using 0.6, 1.0, 1.5 and 2.0 mm core sizes. The arrays are customized for your needs and can be optimized for multi-spectral

Imaging and Analysis
The TDSC performs whole slide imaging using an Aperio ScanScope. Slides can be scanned using 20x or 40x objectives. The digital images are zoomable, can be stored on an
external drive, and can be shared with others on the HUP network.
The TDSC also offers users instruments capable of advanced microscopic image acquisition & analysis. The Vectra and TRIO multispectral slide imaging systems
are available for multi-analyte detection and quantification in a single slide. The inForm analysis software allows users to train the system to analyze large numbers of slides.

The TDSC cuts tissue for molecular analysis using a special clean-cutting protocol. We can macro-dissect tissue and perform nucleic acid extractions. We have a nanodrop,
Bioanalyzer and qPCR machine available for QC and quantification. The TDSC can assist in developing new assays and has worked with the Molecular Diagnostic Lab.
Consultation and Training
The TDSC can help with project planning, tissue selection and experimental design. We can share antibody-specific protocols and provide technical assistance for those doing
IHC in their own labs. Training is required to use the Vectra multi-spectral imaging system and analysis software.

Multispectral slide imaging
The VectraTM system accurately measures protein expressions and morphometric characteristics on whole slides or in distinct tissue regions or TMAs.
Imaged tissue sections can be labeled with immunofluorescent (IF) or immunohistochemical (IHC) stains, or with conventional stains such as H&E and trichrome.
When using IF or IHC stains, one or multiple proteins can be measured on a per tissue, per cell, or per cell compartment (e.g. nuclear, cytoplasmic) basis - even when
signals are spectrally similar, in the same cell compartment or obscured by autofluorescence.
Brightfield and fluorescence imaging
The TRIOTM system can be capture up to three chromogens or up to three fluorophores simultaneously
in a single tissue sample - delivering superior performance over filter-based systems. TRIO also offers multi-labeled per-cell measures of nuclear, cytoplasmic,
and membranous protein expression, making it ideal for monitoring protein signaling pathways and resolving protein co-expression. The system is as straightforward
to use as a point-and-shoot camera - advanced knowledge of multispectral imaging is not needed.
inFormTM advanced image analysis software has the ability to analyze only user-selected regions provides a powerful
solution for performing nuclear, cytoplasmic, and membrane measurements on individual cells.
